Abstrakt     Escherichia coli (E. coli) can cause diarrhoea Virulence factors of strains isolated from humans are well characterised, but little is still known about E. coli strains isolated from diarrhoeic dogs. We analysed 24 bata - haemolytic E. coli strains isolated from the faeces of dogs with diarrhoea at the Utrecht University. The aim of our study was to determine virulence factors and virotypes of above mantioned strains. Each investigated virulence factor (toxins LT, ST, VTI, CNF1 and fimbrial adhesins pap and F17 and the wonfimbrial adhesin intimin) was tested with the Polymerase Chain Reaction (PCR) and hybridisation experiments. The assumption of the presence of a certain virulence factor is based upon multiplying DNA with specific primers in PCR or binding of a specific probe in hybridisation experiment. To confirm the presence of a virulence factor several of the PCR products were cloned and the nucleotide sequence of the cloned fragments was determined using the automatic nucleotide sequence analyser. The obtained sequences were analysed with the program BLAST and an evolutionary distance tree for the papA genes was constructed with the program PHYLIP. The plasmid content of the investigated strains was also examined. For all of the tested strains we demonstrated at least two virulence factors, which can cause or play a role in diarrhoea We thus confirmed that E. coli strains can cause diarrhoea in dogs.
