Abstrakt     We studied the influence of selected substances on ischemic-reperfusion injuries in isolated rat hearts and on isolated porcine coronary arteries. Ischemia was induced either by disconnection of coronary flow or by application of substances that contracted blood vessels and thus evoked myocardial hypoxia. The effects of calcium antagonists mibefradil, selective for T calcium channels, and lacidipin, selective for L calcium channels, were compared. Endothelin-1 (ET-I) and equinatoxin II (EqT II), that both contracted coronary arteries, were also used to evoke cardiac hypoxia. We also studied the influence of tezosentan, a nonselective endothelin antagonist, upon ischemic and hypoxic injuries induced by ET-1 and EqT II in isolated rat heart and isolated porcine coronary arteries; we tried to determine the possible involvement of endothelin system in EqT II activity. Both mibefradil and lacidipin similarly reduced ischemic injuries during reperfusion in isolated rat heart. The effect of mibefradil was similar to that of lacidipin at higher concentration where also negative inotropic effect of mibefradil appeared. ET-1 reduced coronary flow in isolated rat hearts. The transiently increased left ventricular pressure indicates the direct influence of ET-1 on cardiomyocites. Tezosentan efficiently reduced the effects of ET-1 in isolated rat heart. During reperfusion increased lactate dehydrogenase release rate indicated that during ischemia suppressed endothelin system enhanced myocardial injury. In isolated rat heart and in isolated coronary arteries EqT II provoked similar injuries to ET-1. Tezosentan partially reduced the effects of the toxin indicating the possible activation of eridothelin system by EqT II. In isolated coronary arteries both, tezosentan and indomethacin,reduced contractions induced by EqT II, while methylene blue did not. (Abstract truncated at 2000 characters).
