Abstrakt | For detection of fireblight, caused by bacterium Erwinia amylovora (Burrill) Winslow et al., a testing procedure recommended by EPPO (European Plant Protection Organization) is used. It includes: isolation of bacteria by plating on semiselective and non-selective media, serological methods (agglutination and immunofluorescenceJ, biochemical tests and pathogenicify test on immature pear fruits. As an addition to all these laboratory tests we have introduced a molecular biology based method (PCR), which both confirms the Erwinia amylovora (Ea) in a bacterial culture, and enables ro distinguish Ea from other bacteria such as Pseudomonas syringoe pv. syringae, a causal agent of a disease similar to fireblight regarding the symproms, and from other bacteria frequently present on fruit trees. A testing on samples with and without symptoms of first finding of fireblight in Slovenia is presented. | |
Izvleček | Za hrušev ožig, ki ga povzroča bakterija Erwinia amylovora (Burrill) Winslow in sod., uporabljamo postopek laborarorijskega testiranja, ki ga predpisuje organizacija EPPO (European Plant Protection Organization). Podlaga za določitev je izolacija bakterije na semiselektivnih in neselektivnih gojiščih, serološke tehnike (aglutinacijski in imunofluorescenčni test), biokemijski testi in test patogenosti na nezrelih plodovih hrušk. Kot dopolnilo naštetim laboratorijskim testom, smo uvedli molekularno metodo PCR, ki omogoča potrditev izolirane bakterijske kulture Erwinia amylovora in jo razlikuje od bakterije Pseudomonas syringae pv. syringae, povzročiteljice bolezni z zelo podobnimi bolezenskimi znamenji kot hrušev ožig, in od drugih pogostih bakterij sadnega drevja. V prispevku predstavljamo laboratorijsko testiranje na bakterijo Erwinia amylovow iz vzorcev z izraženimi bolezenskimi znamenji in iz latentno okuženih vzorcev prve najdbe ognjevke v Sloveniji. | |
