Abstrakt     Mycoplasma synoviae (MS) abundantly synthesizes hemagglutinin vIhA, which is an immunodominant lipoprotein, associated with its pathogenicity. Recombinations of the 5' end of the vlhA gene with its numerous pseudogenes generate vlhA antigenic variants. In most MS strains hemagglutinin vlhA undergoes posttranslational cleavage into the Nterminal part, a lipoprotein MSPB and C-terminal part, a protein MSPA associated with hemadherence. Recombinations of the vlhA gene have a special potentinal to introduce very diverse sequences of pseudogenes into the region that encodes MSPA. However, there is no data about MSPA sequences of MS populations that changed hemagglutinating (HA) phenotype or MSPA antigenic determinants. In this study we determined coding sequences of the vlhA gene of numerous MS strains. Analyses of the 5'-vlhA sequences revealed new polymorphisms that define new variants (types) of the vlhA gene. Thus, K2559 strain revealed a new type of the vlhA (type F), whereas in other strains new vlhA subtypes (C7 and E3) were found. Characteristic polymorphisms in the 5'-vlhA enable differentiation between MS strains and their analysis was a base for the epidemiologic study of MS strains infecting chicken flocks of the RO comercial line in Slovenia. In those MS strains we found only vlhA of type E. Type vlhA is characterized by a long deletion (`7 nucleotides) in the region encoding proline-rich region of the MSPB protein. However, all MS isolates recovered from tracheas of hens revealed vlhA of subtype E1, whereas other samples relevant to the study eventually showed vlhA of EZ and E3 subtypes. In MS strain ULB925, its subpopulation deriving from its clone KF, revealed 9 variants of the recombined vlhA genes. Four recombined genes revealed at the 5'-vlhA a characteristic codon change (403GCT->AAA) indicating site-specific recombination. (Abstract trunacted at 2000 characters).
