Abstrakt | Nonsteroidal anti-inflammatory drugs (NSAIDs) have been used over many decadesfor treatment of inflammation and pain. Due to prostaglandin inhibition, these drugs cause gastrointestinal adverse effects, disturbances of kidney function and blood coagulation. Clinically significant gastrointestinal adverse effects are ulcers, hemorrhages and perforation. Nonsteroidal anti-inflammatory drugs differ mainly in their potency of inhibition of two cyclooxygenase isoforms, COX-1 and COX-2. The newer class ofthese drugs, coxibs, have a less potent effect on prostaglandin synthesis and cause fewer gastrointestinal adverse effects. The safe use of NSAIDs is based on careful selection of patients, correct dosage and intake of the prescribed drug as well as effective protection of patients with risk factors. | |
