Abstract		The cardiovascular effects of amlodipine and nicardipine, dihydropiridine calcium channel blockers, have been studied on the isolated thoracic aorta of the rabbit without endothelium and on the isolated spontaneously beating guineapig atria. Changes of tension were monitored isometrically. Amlodipine (IC50 = 1.23x10-8M, 3rd hour) and nicardipine (IC50 = 3.24x10-9M, 2nd hour) caused relaxation on the rabbit thoracic aorta, depolarized by 40 mM KCl. Equimolar concentration of amlodipine or nicardipine (3x10-8M) produced in 10th minute, qualitative and quantitative different effect on the guinea pig atria; amlodipine slightly positive inotropic, nicardipine slightly negative inotropic, both compounds caused negative chronotropic effect, however, amlodipine produced significantly smaller negative chronotropic effect than nicardipine (P less th. 0.05).
