Abstract | The first human tumour virus, virus Epstein-Barr, was discovered in 1964 in Burkitt's lymphoma cells. Oncogenic viruses are important for two main reasons. Firstly, approximately 15% of human cancer incidence can be attributed to a viral infection. Secondly, oncogenic viruses are analogous to other mutagenic carcinogens, being capable of initiating a chain of two or more events possibly leading to malignancy. Today it is known that many retroviruses produce tumours in domestic animals, and one has been implicated as a cause of human leukemia. The hepatitis B virus, flaviviruses, herpes viruses, especially the Epstein-Barr virus and herpes simplex virus 2, and several papillomaviruses are also implicated in cancer in humans. The greatest prospect for significantly reducing the frequency of cancer in humans is through immunisation against infection with oncogenic viruses. | |
Summary | Prvi humani onkogeni virus, virus Epstein-Barr, so odkrili leta 1964 v celicah Burkittovega limfoma. Onkogeni virusi so pomembni, ker so vpleteni v nastanek 15% rakavih obolenj pri človeku in ker so sposobni aktivirati verigo dogodkov, ki lahko podobno kot drugi karcinogeni dejavniki vodijo v maligne spremembe. Danes vemo, da retrovirusi ne povzročajo rakavih sprememb samo pri živalih, ampak tudi pri človeku. Človeške onkogene viruse uvrščamo v družine papilomavirusov, herpesvirusov, flavivirusov, retrovirusov in hepadnavirusov. Med herpesvirusi sta zlasti pomembna virus Epstein-Barr in virus herpesa simpleksa 2. Vse več pa je tudi dokazov o onkogenem delovanju virusa citomegalije in človeškega herpesvirusa 6 in 8. Prihodnost v boju proti malignim spremembam, ki jih povzročajo virusi, temelji predvsem na imunizaciji, s katero bi ljudi in živali zaščitili pred onkogenimi virusi. | |
