Abstract     The objective of this study was to examine the organization of the actin cytoskeleton in human embryonic kidney cells (HEK 293 cells) that had been stably transfected with a thyrotropin-releasing hormone (TRH) receptor. The emphasis was on its rearrangement induced by the activation of G-protein coupled receptors (GPCR) exemplified by the TRH receptor. Morphometric cell analysis was performed at the light microscope level. Distribution of fluorescently-labelled actin filaments was monitored by confocal microscopy. Agonist stimulation of cells expressin.g the TRH receptor resulted in an increase in their surface area and the reorganization of their actin network. This agonist-induced effect can be abolished by the concomitant treatment of the cells with cytochalasin D. These results demonstrate that an agonist-induced activation of the TRH receptor, a member of the GPCR family, causes a change in a cell's morphology that probably correlates with the rearrangement of the actin cytoskeleton.
Summary     Z delom smo želeli proučiti razporeditev aktinskih nitk v in vitro gojenih humanih ledvičnih celicah HEK 293, trajno preoblikovanih z receptorjem za tiroliberin (TRH). Poudarek je bil na prerazporeditvi mreže aktinskih nitk po aktivaciji s proteinom G združenih receptorjev (GPCR), ponazorjenih z receptorjem za TRH. Morfometrično analizo celic smo opravili s svetlobno mikroskopijo. Razporeditev fluorescenčno označenih aktinskih nitk pa smo spremljali s pomočjo konfokalne mikroskopije. Stimulacija celic, ki trajno izražajo receptor TRH, z agonistom TRH je povzročila povečanje površine tretiranih celic ter preoblikovanje mreže aktinskih nitk. Vpliv agonista lahko preprečimo s sočasno obdelavo celic s citohalazinom D. Dobljeni rezultati kažejo, da aktivacija receptorja TRH, predstavnika družine GPCR, povzroči spremembo oblike celice, ki najverjetneje sovpada s preoblikovanjem mreže aktinskih nitk.
