Abstract     Electroporation, i. e. application of electric pulses to the cells or tissues, induces under suitable conditions reversible permeabilization of cell membrane, thus facilitating entry into the cells of nonpermeant or poorly permeant molecules, including chemotherapeutic drugs. Some chemotherapeutic drugs such as bleomycin and cisplatin are potent cytotoxic agents but are poorly transferred across the cell membrane due to their chemical nature, molecular weight and the deficiency of transport mechanisms. Treatment of cells or tumours with the application of electric pulses and chemotherapy is called electrochemotherapy. In addition to cytotoxic effect, bleomycin and cisplatin have also radiosensitizing effect. Both are used in the clinic for treatment of malignant tumours in the combination with other chemotherapeutic drugs or irradiation. The purpose of the Doctoral dissertation was to evaluate radiosensitizing effect of bleomycin and cisplatin after application of electric pulses (electroporation) on LPB sarcoma tumour cells and tumours. We hypothesized that radiosensitizing effect of bleomycin and cisplatin is increased due to the localized and increased uptake of the drugs into the LPB tumour cells and tumours by electroporation. The effect of electroporation on radiosensitizing effect of bleomycin and cisplatin on LPB sarcoma tumour model was evaluated in vitro and in vivo. The effect of electroporation on radiosensitizing effect of bleomycin and cisplatin was determined at different drug and irradiation doses by clonogenic assay. The radiosensitizing effect of bleomycin and cisplatin on LPB cells was increased by electroporation. This combined treatment was more effective when using bleomycin, compared to cisplatin. Electrochemotherapy with bleomycin potentiated cytotoxicity of irradiation by a factor of 1.53 and by factor of 1.35 in the case of electrochemotherapy with cisplatin.
