Title     Plasma fibrinogen determination by different methods

Abstract     Plasma fibrinogen determination is a clinically important assay, for which many methods are currently used. We have compared the analytical performances of five methods: biuret, ACL (Instrumentation Laboratory), Fibrin-timer (Behring), Chromo-timer (Behring) and immunonephelometry. Good within-run reproducibility of each method was obtained (CVs 1.2-4.4 per cent). The largest day-to-day variation was found for immunonephelometry (5.6-8.7 per cent), whereas other four assays had their day-to-day CVs ranging from 2.5 to 5.3 per cent. The sensitivities of ACL, Fibrin-timer and immunonephelometry corresponded to the lower limit of instruments measuring range, i.e. 0.3, 0.3 and 0.25 g 1 power -1, respectively. Sensitivity of 0.5 g 1 power -1 was obtained for the Chromo-timer and 0.7 g 1 power -1 for the biuret method. Inter-method comparison showed that biuret and ACL correlated well with the Fibrin-timer method as reference (r = 0.976 and 0.986, respectively). When the results of the Chromo-timer and Fibrin-timer methods were compared, a high correlation coefficient was obtained (r = 0.978), but with systemic difference as large as 15.1 per cent, Chromo-timer yielding lower results.
