Abstract     Human neutrophil antigens (HNA) are clinically important. Alloantibodies directed against them can cause alloimmune and isoimmune neutropenias in newborns, adverse transfusion reactions, such as febrile nonhaemolytic transfusion reaction and TRALI (Transfusion-related acute lung injury), refractoriness to granulocyte transfusions, prolonged neutropenia following stem cells transplantations and drug induced neutropenias. Autoantibodies can cause primary and secondary autoimmune neutropenias. The aim of the study was to determine antigen and gene frequencies of HNA-1a, -1b, -1c, -2a, -3a and -4a as well as the frequency of HNA-1 "null" phenotype in the Slovene population and to compare them with the data from other populations. We wanted to find out the haplotype, in which FCGR3B*3 is inherited in the Slovene population. The aim was also to get new members typed in HNA for the register of typed donors. HNA were typed in random, healthy, unrelated blood donors and employees of the Blood Transfusion Centre of Slovenia. The frequencies of HNA-1a, -1b, -1c and HNA-1 "null" were determined by genotyping of FCGR3B*1, FCGR3B*2 and FCGR3B*3 from genome DNA by PCRSSP in 318 persons. The inheritance of FCGR3B*3 was studied in three FCGR3B*(3+) persons coming from three different families in which eight relatives were genotyped. The frequencies of HNA-2a, -3a and -4a were determined serologically, with antigen specific human sera. HNA-2a was typed in 194 and HNA-4a in 167 persons with a granulocyte immunofluorescence test. HNA-3a was determined in 194 persons with a granulocyte agglutination test. The absence of HNA-2a was confirmed serologically with MAIGA (Monoclonal antibody-specific immobilisation of granulocyte antigens). In the Slovene population the antigen frequencies of HNA-1a, HNA-1b and HNA-1c are 0.642, 0.871 and 0.075 and their gene frequencies are 0.385, 0.615 and 0.037. The phenotype frequency of HNA-1 "null" is < 0.003. (Abstract truncated at 2000 characters).
