Abstract     Tumor necrosis factor-a (TNF-alpha) is a pleiotropic cytokine which, as a part of complex cytokine system, controls the synthesis and expression of other cytokines and their receptors through the activation of cytokine cascades. The aim of our study was to test the effects of TNF-alpha on the secretion of interleukin-6 (IL-6) from cultured human muscle. We studied these effects before and after the fusion of myoblasts into multinucleated myotubes. Cultures, prepared from satellite cells isolated from the muscle pieces routinely discarded at some orthopaedic operations were treated with TNF-alpha at three different concentrations. We found that TNF-alpha increases the IL-6 secretion in a concentration dependent manner. Quantitatively, secretion from myotubes exceeded the secretion from myoblasts indicating that secretion is differentiation stage-dependent. The presence of TNF-alpha receptor was confirmed by Western blot. Apoptosis, followed by DNA fragmentation, remained unchanged. Our results support the concept that skeletal muscles actively participate in cytokine signalling in critical clinical conditions.
Summary     Dejavnik tumorske nekroze-a (TNF-alfa) je pleiotropni citokin, ki kot del kompleksnega citokinskega sistema uravnava sintezo in ekspresijo drugih citokinov in njihovih receptorjev prek aktivacije citokinskih kaskad. Namen naše naloge je bil proučiti učinek TNF-alfa na izločanje interlevkina-6 (IL-6) iz človeške mišice v kulturi, in sicer na stopnjah pred fuzijo mioblastov v mišične cevčice in po njej. Kulturam, pripravljenim iz satelitskih celic, ki smo jih izolirali iz koščkov skeletnih mišic, ki se po doktrini odstranijo in zavržejo pri nekaterih ortopedskih operacijah, smo dajali TNF-alfa v treh različnih koncentracijah. Ugotovili smo, da TNF-alfa koncentracijsko odvisno pospešuje izplavljanje IL-6. Količine izplavljenega IL-6 so bile v primeru mišičnih cevčic večje kot v primeru mioblastov, kar kaže na odvisnost učinka TNF-alfa od stopnje mišične diferenciacije. Prisotnost receptorjev za TNF-alfa smo dokazali s tehniko Western blot. Morebitne pospešene apoptoze, ki smo jo ugotavljali s fragmentacijo DNA, nismo ugotovili. Naši rezultati podpirajo domnevo o aktivni vlogi skeletne mišice pri citokinskem signaliziranju v kritičnih kliničnih stanjih.
