Abstract     Antibodies against beta2-glycoprotein I (anti-beta2-GPI) are among the most commonly detected subsets of antiphospholipid antibodies. Nevertheless, numerous questions about methodologies of anti-beta2-GPI detection considering rational laboratory diagnostics, pathogenic mechanisms and clinical associations are still matters of intensive studies. Answers to these questions will refine their diagnostic meaning. We hypothesize that avidity of anti-beta2-GPI importantly influences their pathogenic potential and consequently also clinical relevance of laboratory findings. Affinity and cationic exchange chromatography were used to isolate beta2-GPI, polyacrylamide gel electrophoresis, N-terminal sequencing and immunochemical methods for the characterization of isolated proteins, and chaotropic enzyme-linked immunosorbent assay for the determination of avidity of IgG anti-beta2-GPI.
