Abstract     The aim of this study was to evaluate electrogenetherapy with p53wt alone or combined with cisplatin in two colorectal (HT29 and LoVo) and two prostatic (PC3 and Du145) carcinoma cell lines with different p53 statuses. Electrogenetherapy with p53wt was dependent on the p53 status of the cell lines used. Electrogenetherapy was the most effective in PC-3 (p53 null) and Du145 (p53mt) cells and to a much lesser extent in LoVo cells (p53wt). An exception was the HT-29 cell line with an overexpressed mutated p53, in which electrogenetherapy with p53wt was the least effective. Sensitivity of the cell lines to cisplatin was independent of the p53 status. Furthermore, the presence of exogenous p53 due to electrogenetherapy did not enhance cisplatin cytotoxicity, since the combination of these therapies resulted in an additive cytotoxic effect.
Summary     Namen naše študije je bil določiti učinek elektrogenske terapije s p53wt kot samostojne terapije ali v kombinaciji s cisplatinom na preživetje človeških celičnih linij karcinoma debelega črevesa (HT29 in LoVo) ter karcinoma prostate (Du145, PC3). Elektrogenska terapija s p53wt je bila odvisna od celičnega statusa proteina p53. Najbolj je bila učinkovita na celičnih linijah PC3 (brez p53) in Du145 (mutiran p53), manj pa na celični liniji LoVo (p53 wt). Izjema je bila celična linija HT29, ki ima povečano izražanje mutiranega p53, kjer je bila elektrogenska terapija najmanj učinkovita. Status proteina p53 v različnih celicah ni vplival na citotoksičnost cisplatina. Prav tako tudi prisotnost eksogenega proteina p53 divjega tipa zaradi elektrogenske terapije ni vplivala na povečanje citotoksičnosti cisplatina. Součinkovanje elektrogenske terapije in cisplatina se je namreč pri vseh celičnih linijah seštela.
