Abstract     Background. A change of erythrocyte shape is necessary during blood circulation. Deformability of erythrocytes is hence one of their basic properties. Aim. Research project tried to elucidate role of submembrane protein skeleton in mechanical stability of erythrocytes. Our hypothesis suggested that change of mechanical properties of protein skeleton should result in a change of deformabiltiy of membrane of erythrocytes. Materials and methods. We have traced the properties affecting deformability of erythrocytes by observing changes of shape of erythrocytes due to variations in external factors. We induced change of shape of erythrocytes with sodium salicylate, which transforms biconcave disc (physiological shape of erythrocytes) into echinocyte - sphere with numerous membrane spicules. We applied diamide to strengthen submembraneous protein skeleton and again observed changes of shape due to sodium salicylate. Results. Results suggest diamide lowers deformability of the membrane. Decrease of deformabiltiy depends on concentration of diamide used. Conclusions. Deformabilty of membrane depends on mechanical characteristics of submembrane protein skeleton. Decrease of deformability is a result of higher skeleton strength.
