Abstract     The focused ion beam (FIB) / SEM system was used to prepare cross-sections of selected regions of digestive gland epithelium of Porcellio scaber (Isopoda, Crustacea) for scanning electron microscopy. Application of a FIB / SEM system for morphological studies has advantages over conventional microscopy techniques because of: in situ site-specific 2D-sectioning and imaging of subsurface microstructures; no need to embed the sample prior to sectioning; and a possibility of using a wide range of magnifications while imaging the same sample. The FIB / SEM system is in the early stage of application in life sciences and appears to be a technique of choice for 3D-observations at the organism, organ, tissue and (sub)cellular levels.
