Abstract     Unravelling the molecular mechanisms of gastric carcinogenesis is one of the major challenges in cancer genomics, because gastric cancer is a very heterogeneous model disease and although much has been learned about the DNA changes that eventually lead to its development, the detailed mechanisms still remain unclear. Malignant growth of cancer cells is a result of complex genetic and epigenetic changes, which occur during multistep process of gastric cancer pathogenesis. Although the functional characterization of individual genes sheds the light on their true relevance in cancer development, it is necessary to use genome-wide screening techniques to isolate a spectrum of differentially expressed genes in tumor tissues, especially for understanding complex cancers that are the consequence of changes in many different genes. Screening for differentially expressed genes is a straightforward approach to elucidate molecular basis of a disease and comparing molecular signatures of changes that occur in tumor tissues at different stages of development holds a great promise to advance our understanding and treatment of gastric cancer. The purpose of this study was to identify gene expression profiles in tissues of gastric cancer and in biopsies of gastric precancerous lesions by constructing a cDNA library with suppressive subtraction hybridization (SSH). Furthermore, we wanted to find out if there exist differences in gene expression between individual patients. Data analyses of differentially expressed genes showed differences in gene expression in tumor tissues compared to corresponding normal mucosa and also differences between precancerous lesions compared to gastric tissues.
