Abstract     Bacterial blight of grapevine has been occurred in Primorska winegrowing region since the late fifties of the previous century, while its causer Xylophilus ampelinus (Panagopoulus) Willems et al. has been confirmed by laboratory means only recently. In the present article a chronology of its appearance and current distribution in Slovenia are presented, as well as symptoms and the results of the monitoring in 2003 discussed. Bacterium Xylophilus ampelinus (Pananogopoulos, 1969) comb nov. (Willems et al., 1987) is a causative agent of bacterial blight of grapevine. Laboratory detection following Koch's postulates is particularly demanding with these bacteria due to their uneven distribution in plants and slow growth on artificial media. A sample of grapevine (Vitis vinifera L.) cv. Rebula from a vineyard in Primorska region, exhibiting characteristic angular lesions on leaves was recieved for testing in 2002. An isolate was obtained and confirmed as X ampelinus using biochemical and nutritional tests, fatty acid analysis, immuno-fluorescence, nested PCR, partial sequencing of the 16S rRNA gene and pathogenicity test. The pathogenicity test was performed on grapevine cv. Rebula grown in tissue culture. Symptom development in the vineyard was followed and monitoring of X. ampelinus performed for two subsequent years.
Summary     Xylophilus ampelinus (Pananogopoulos, 1969) comb nov. (Willems et al., 1987) povzroča bakterijski ožig vinske trte. Laboratorijsko določanje bakterij, ki skuša slediti Kochovim postulatom, je pri tej bakteriji še posebej težavno zaradi njene neenakomerne razporeditve po rastlini ter počasne rasti na gojiščih. Iz vzorca vinske trte (Vitis vinifera L.) sorte Rebula odvzetega na področju Primorske v letu 2002, ki je kazal za to bolezen značilne pege na listih, smo izolirali kolonije po morfologiji podobne kolonijam kontrolne bakterije. Z uporabo biokemijskih in nutritivnih testov, profilom maščobnih kislin, imunofluorescenco, vgnezditveno PCR reakcijo, določanjem zaporedja nukleotidov dela gena za 16S rRNA ter testom patogenosti na rastlinah vinske trte gojene v tkivni kulturi smo potrdili, da gre res za X ampelinus. Pojavljanje znamenj bolezni ter učinkovitost diagnostičnih metod smo še nadaljnji dve leti spremljali v okvim posebnega nadzora bakterijskega ožiga vinske trte v koordinaciji Fitosanitarne uprave Republike Slovenije.
