Abstract     Gene expression analysis of glomerular disease from routine renal tissue biopsies requires the specific isolation of the target structure, as well as quantification of the levels of genes from a very low amount of template. Laser microdissection has recently been established as a reliable method for isolating morphologically distinctive tissue regions with no or very low contamination with surrounding tissue components. Real time RT-PCR combines excellent sensitivity with quantification by the direct determination of amplicon numbers after each PCR cycle. We studied the gene expression of VCAM-1, fractalkine, IL-6 and bax in laser microdissected glomeruli from 27 needle biopsies of lupus nephritis patients. We found higher values of gene expression in lupus cases compared to controls, although the relative expression values showed considerable variation among cases. Our study demonstrates that the introduction of molecular diagnostics to renal biopsies could result in improved prognostic information and more relevant selection of available therapeutic strategies.
