Abstract     Thanks to their high therapeutic efficiency, corticosteroids are among the most widely prescribed classes of drugs. Their use is, however, restricted by serious side effects. The actions of corticosteroid medications as well as their side effects can be better understood in the context of the physiological role of their endogenous counterparts, which act along the same pathways as their synthetic analogues. The homeostatic mechanisms controlling the release of corticosteroid hormones are first described. This is followed by a description of their effects and finally, the basic molecular mechanisms underlying corticoid effects on the transcription of genesis discussed.
Summary     Kortikosteroidi sodijo med zdravila, ki jih zdravniki najpogosteje predpisujejo. Odlikuje jih visoka terapevtska učinkovitost, rabo pa omejujejo resni stranski učinki. Oboje je laže razumeti, če poznamo fiziološko vlogo endogenih steroidnih hormonov, ki se izločajo iz skorje nadledvičnice in ki delujejo po istih poteh in mehanizmih kot njihovi sintetični analogi. V tem poglavju najprej opišemo homeostatske mehanizme, ki urejajo izločanje kortikosteroidnih hormonov, sledi opis njihovih učinkov, v zadnjem delu pa razložimo osnovne molekularne mehanizme, prek katerih kortikosteroidi prožijo svoje učinke na ravni genskega prepisovanja.
