Abstract     Background. Remodeling of alveolar bone and the periodontal ligament is the key process in orthodontic tooth movement. It consists of tissue formation and resorption. On the pressure side of the loaded tooth resorption of alveolar bone and periodontal ligament takes place. On the tension side new bone is formed and the periodontal ligament remains intact. Many chemical messengers are involved in the process of remolding of the alveolar bone and the periodontal ligament. One of the chemical messengers could be endothelin. The role of endothelin in the process of orthodontic tooth movement has not been explained yet. The only thing known is that tesosentan, a nonselective endothelin antagonists (ETA in ETB), enhances orthodontic tooth movement in rats. AIM. The aim of the study is to determine a more precise role of the endothelin system in orthodontic tooth movement. Therefore we used TBC 3214, a selective ETA endothelin antagonist. Hypothesis. We suspect that the endothelin system is included in the process of tooth movement and that TBC 3214, a selective ETA endothelin antagonist, will influence the magnitude of tooth movement. Methods. The study was performed on male Wistar rats divided into 3 groups (n=10). The first group was applied a tractive coil spring and was daily treated with tesosentan. The second group was also applied a tractive coil spring and was daily treated with physiologic solution. Third group performed as a control group and was only treated with physiologic solution daily. The tractive coil spring was applied between the upper left first molar and upper left incisor. Measurements between the mentioned teeth were taken with a digitronic caliper (precision 0,01 mm) on days 0, 7, 14, 21, 24, 27, 32, 37 and 40. Graph Pad Prism 4.00 and Two way Anova were used for statistical analysis. Differences in distance between the mentioned teeth were calculated to determine the amount of tooth movement.
