Abstract     Sulphidoleukotrienes (sLT) are important mediators of allergic and pseudoallergic diseases. We studied the ability of basophils of patients with allergic and pseudoallergic diseases to release sLT. Leukocytes of 18 patients with Hymenoptera venom allergy (10 atopics, 8 nonatopics), 14 patients with chronic urticaria (CU) and 11 healthy volunteers were in-vitro stimulated with antibodies against high affinity IgE receptors (anti-FcsRl) and ionomycin and amount of released sLT was measured by ELISA assay. Spontaneous and anti FcsRl induced release of sLT was diminished in patients with CU. lonomycine induced release of sLT in CU patients was similar as in healthy controls but it was significantly higher in patients with allergic diseases. Anti FcERI induced release of sLT from leukocytes of atopic allergic patients was significantly higher than release of sLT from leukocytes of healthy controls and nonatopic allergic patients. We specutate, that weaker response to antiFcsRl and normal response to ionomycine in basophils of CU patients might be due to lower number of available FcsRl (lower number or occupancy with blocking autoantibodies). Higher sLT releasability of leukocytes of allergic patients after stimulation with ionomycine might be a mechanism responsible for development of overt allergic disease in patients having specific IgE antibodies.
