Abstract     Background. Moderate red wine consumption reduces the incidence of cardiovascular diseases despite some other risk factors (French Paradox). Beside alcohol, red wine contains also polyphenolic compounds which protect cardiovascular system. Aim. We compared effects of red wine free of ethanol (deEtOH) and deEtOH wine free of polyphenols on relaxation of isolated rings of porcine left coronary artery (LAD), and tried to determine polyphenolic compound of wine with most relaxant effects on LAD and on rat aorta (RA). Hypotheses. We supposed: (1) deEtOH red wine inhibits with KCl induced contraction of LAD; (2) deEtOH wine free of polyphenols has a relaxant effect on LAD and (3) different polyphenolic compounds relax LAD and RA to different degrees. Methods. Isolated LAD and RA rings (3-4 mm wide) were placed in organ baths filled with Krebs-Henseleit solution and oxygenated with 95% 02 and 5% C02. In experiments in LAD rings with concominant cyclooxygense (COX) and nitric oxide synthase (NOS) inhibition we evaluated contractions induced with endothelin (ET-1) or with U46619 (a tromboxane agonist). We also evaluated contractions induced with ET-1 in LAD rings without inhibition of COX and NOS. In the second series of experiments we evaluated KCl (10-90 mM) induced contractions of LAD before and after 30-minutes incubation in deEtOH red wine. The relaxation of LAD rings, precontracted with ET-1, by different concentrations of red wine free of polyphenols and of wine that contained polyphenols, was also determined. LAD rings precontracted with ET-1 and RA rings precontracted with norepinephrine were relaxed with different polyphenolic compounds of red wine and effects were compared. The effect of catechin on KCl induced contraction of LAD rings was also evaluated. (Abstract truncated at 2000 characters)
