Abstract     The transplantation of the hematopoietic stem cells (HSC) has become the standard treatment for several malignant and genetic diseases. The CD34 antigen is the marker present on the membrane surface of HSC. HSC can be collected from the bone marrow, peripheral blood and from umbilical cord blood. Blood cell separators are used to collect HSC from the peripheral venous blood. Before collection the small number of circulating HSC have to be increased using the hematopoietic growth factors, chemotherapeutic agents or their combination in order to mobilize HSC from the bone marrow into the blood. During the mobilization using growth factors, the levels of gelatinase B - matrix metalloproteinase -9 (MMP-9) are increased. MMP-9 have a documented role in the not yet fully understood mobilization mechanism. Previous studies have shown additional transmigration and egress of CD34+ cells into the circulation during the collection by leukocytaphereses. Our hypothesis is, that this additional transmigration of CD34+ cells during the collection can be related to MMP-9 enzymatic activity. There are also poor mobilizers, particularly among the autologous donors, with a low number of mobilized CD34+ cells who need higher doses of growth factors and more collection procedures or who fail to collect a sufficient numer of CD34+ cells for transplantation. Because of the hypothetical association between MMP-9 concentration before collection and the number of collected cells, we expected that the number of mobilized or collected CD34+ cells can be predicted from the known concentration of MMP-9 before collection. There are no data in literature about the changes of MMP-9 levels during the collection of HSC by leukocytapheresis and its association with the collected CD34+ cells.
