Abstract     Background: Brain-derived neurotrophic factor (BDNF) is an important neurotrophin larown to provide neurotrophic support by promoting neuronal survival and differentiation during development, by mediating specific forms of structural and functional plasticity in the adult brain and by providing neuroprotection after nerve injury. BDNF is produced by neurons and by non-neuronal cells - astrocytes, that represent an important local source of trophic support in normal and diseased brain. Our previous studies demonstrated that asirocytic BDNF synthesis is susceptible (to varying degrees) to stimulation by different pharmacologicaly active substances (neurotransmitters, neuromodulators, steroid hormones). Astrocytes are part of the tripartite synapse and express various neurotransmitter receptors, therefore the present study was focused on the regulatory effects of monoamine neurotransmitters on the synthesis of BDNF protein in astrocytes. Aim: In the present work I examined dose-dependency and short-term kinetics of the regulatory effects of monoamine neurotransmitters noradrenaline (NA), serotonin (5-HT) and dopamine (DA) on the synthesis of BDNF protein in cortical neonatal rat astrocytes and, in addition, investigated the involvement of the specific adrenergic receptor subtypes in the regulation of astrocytic BDNF by NA. Hypothesis: BDNF synthesis in asttocytes is dependent on monoaminergic neuronal activity, and in vitro is dependent on the concentration and time of exposure to monoaminergic neurotransmitters (noradrenaline, serotonin, dopamine). Stimulation of BDNF synthesis with monoaminergic neurotransmitters is a receptor mediated process (noradrenaline stimulates BDNF synthesis via adrenergic receptors).
