Abstract		The inhibitory effect on the contraction of the thoracic aorta of the rabbit and negative inotropic and chronotropic potencies of the two stereoisomers of nicardipine (NC) on the quinea-pig atria were compared with racemic mixture. Changes in tension were monitored isometrically on the spiral strip from rabbit thoracic aorta (potassium chloride induced contractions) and on the isolated guinea-pig atria (spontaneous activity). In both cases equimolar concentrations of nicardipine chloride were used. The strongest effect on the thoracic aorta of the rabbit was achieved with NC(+-), followed by NC(+) and NC(-) forms. The strongest effect on the spontaneous action of isolated guinea-pig atria in the sense of negative inotropic potency was also achieved with NC(+-), followed by NC(+) and NC(-). The negative chronotropic effect was about the same with NC(+) and NC(+-), and slightly less potent with NC(-).
