Abstract     The duodenal tissue of thirty-six (36) healthy fatteners was used for this research. The animals were divided in five groups that differed according to weight and husband. The mean animal weight was 112.0 kg. The duodenal tissue was fixed with Bouin's and Carnoy's fixative solution, with 4 per cent basic lead acetate in water, with 10 per cent and with 4 per cent neutralized formalin. The tissue slices of 7čm were stained with Toluidine blue at H 4.2, with hematoxylin and eosin, with Masson's threechrome staining, Schiff's reagent and with a combination of Schiff's reagent and Alcian blue. Volume, surface and specific surface density of tissue mast cells in the pig duodenum were determined stereologically. The results were statisticall interpreted according to fixatives and groups of animals. The mast cells were specifically stained with Toluidine blue at pH 4.2. Metachromasia was well marked. Mast cells were determined in the lamina propria, submucosa Brunner's glands and tunica muscularis. They were not found in the lamina epithelialis. The mast cells were PAS negative. The established values for volume density differed significantly in correlation with fixatives. Basic lead acetate and Bouin's fixative soluhon preserved mast cells in the duodenal wall similarly well. Carnoy's fixative solution showed less mast cells and was comparable with 10 per cent formalin in the case of tissue submucosa and tunica muscularis. 10 per cent formalin gave better results than 4 per cent formalin yet neither of them was suitable for demonstrating the mast cells in the mucosa of the pig duodenum. After Bouin's fixative being used, it has been determined that mast cells occupy the largest portion of tissue volume in the mucosa (1.62 +- 0.10 per cent ), less in the submucosa (1.50 +- 0.11 per cent ), Brunner's glands (0.92 +- 0.07 per cent ) and tunica muscularis (0.52--č 0.06 per cent ).(trunc.)
