Abstract     Colorectal cancer carcinogenesis is a complex, long-lasting multistep process. Despite intense research in this field there is still no objective marker which would help us in predicting clinical course of bowel dysplasia and identify patients with the highest risk for colorectal cancer development. Patients with different polyposis syndromes, including classical FAP (Familial Adenomatous Polyposis), attenuated FAP, MUTYH-associated polyposis (MAP), Peutz-Jeghers syndrome, juvenile polyposis syndrome, Cowden syndrome, hereditary mixed polyposis syndrome (HMPS) and hyperplastic polyposis syndrome have significantly increased risk for different types of cancer. In this paper the authors present the characteristics of Peutz-Jeghers syndrome, a condition with increased risk of malignancy for the patient and the patient's relatives, their molecular and genetic features and associations with carcinogenesis.
Summary     Karcinogeneza raka debelega črevesa in danke je večstopenjski proces, ki je dolgotrajen in zapleten. Žal kljub intenzivnim raziskavam na tem področju še nimamo objektivnih kazalcev, s katerimi bi lahko napovedali preobrazbo sluznice črevesa in bi nam pomagali odkriti najbolj ogrožene bolnike za rakavo bolezen. Bolniki s polipoznimi sindromi, kot so FAP (Familial Adenomatous Polyposis) ali oslabljena oblika tega sindroma, MUTYH-pridružena polipoza (MAP), Peutz-Jegherjev ali Cowdenov sindrom, oblika juvenilne polipoze, hereditarni mešani polipozni sindrom (HMPS)in hiperplastični polipozni sindrom imajo povečano tveganje za vznikanje različnih vrst raka. V članku avtorja predstavita značilnosti Peutz-Jegherjevega sindroma, stanja, pri katerem je pomembno povečano tveganje za vznikanje različnih oblik raka pri bolnikih in njihovih svojcih, molekularnogenetske osnove te bolezni in povezave s karcinogenezo.
