Abstract     In our present work we study the effect of physicochemical properties of some compounds with different polarity on histamine and serotonin secretion from isolated rat peritoneal mast cells. The experiments can be devided into two groups: 1) the study of the effect of cardiotonic glycosides having different polarity on histamine secretion. 2) the study of the effect of polarity and disociation of some basic compaunds on histamine and serotonin secretion. The results of our study show: 1) The preincubation of mast cells with watersoluble cardiotonic glycosides (ouabain, digoxine) increase histamine secretion. The increase is due to the inhibition of Na+K+-ATPase activity and depends on the presence of extracelular Na+ and Ca2+ ions. Liposoluble glycosides and their genines have a duel effect on the secretion: - the inhibition of Na+K+-ATPase increases histamine release, - an intracellular action of liposoluble glycosides on microfilaments leads to a decrease of the secretion. 2) The effectiveness of basic secretagogues depends on their physico-chemical properties (basic and lipophilic characteristics of the compound). The binding of protonated amino groups of basic compounds to negatively charged structures on the surface of the cell membrane could be a prerequisite for the action of these compounds. After that the lipophilic part of basic compound penetrates through mast cell membrane. This leads to the subsequent interaction of basic compound with GTP-binding protein which is an important step in the process of the exocytotic mehanism.
