Abstract     Alzheimer's disease is patomorphologically characterised by two types of lesions, i.e. neurofibrillary tangles and senile plaques. It has been reported, that the neurofibrillary tangles are the consequence of neuronal degeneration an remain in the extracellular space after neuronal cell death. Senile plaques are comprised of extracellular amyloid deposits in the centre of the plaque, surrounded by degenerating or dystrophic neurites. Despite many studies the relationship betwee neurofibrillary tangles, dystrophic neurites and extracellular amyloid beta deposits remains to be established. We anticipated that the neurofibrillary pathology and the amyloid deposition in Alzheimer's disease are related processes and may represent different stages in the course of disease. We tried to confirm the hypothesis that beta precursor protein accumulates in the dystrophic neurites of senile plaques and is metabolised to amyloid beta in the extracellular space. We tested this hypothesis by analysing the ultrastructural and antigenic characteristics of the dystrophic neurites, the neurofibrillary tangles and the senile plaques. In the study, 3 biopsy and 4 autopsy specimen of brain tissue of the patients fulfilling internationally confirmed criteria for diagnosis of Alzheimer's disease were included. In the biopsy tissue the ultrastructure of senile plaques was analysed. The autopsy tissue was examined by the light and electron microscopic methods of double-labelling. The specific antisera against various components of senile plaques and neurofibrillary tangles - antiserum against protein tau, ubiquitin, beta precursor protein, amyloid beta, basic fibroblast growth factor (for demonstration of heparan sulphate proteoglycans), synaptophysin, cathepsin, glial fibrillary acid protein and neurofilaments were used.(trunc.)
