Abstract     Mucoadhesive oral films containing metronidazole for the teraphy of mouth diseases were designed by the casting procedure, using aqueous dispersion of the drug, PMMA-Na polymer and plasticizer, and subsequent drying. The films are structured as non porous homogeneous matrices. After hydration, they undergo the process of bioadhesion in vivo, retaining at the site of application three to five hours. The hydration profile of the film in vitro is afected by the drying procedure, and the rate of hydration by the plasticizer and thickness. A wide range of drug release in vitro is reached by varying these parameters. Metronidazole release rates are controlled by dissolution kinetics. An appropriate analytical approach was developed for the evaluation of the drug bioavailability in saliva. Evaluation results have shown that the application of optimally designed films assures concentration of metronidazole above minimal inhibitory concentration for five hours.
