Abstract     Candida albicans je eden od številnih saprofitnih gliv na sluznicah človeka. Najdemo jo v ustih, v dihalih, prebavilih in v vagini pri 25% povsem zdravih ljudi. Je najpogostejša gliva, ki je odgovorna za večino lokaliziranih glivičnih infekcij pri ljudeh. Bolniki z zmanjšano imunsko odpornostjo, onkološki bolniki in bolniki okuženi z virusom HIV so še posebej ogroženi za razvoj glive Candida albicans. Metode V raziskavi smo pri različnih koncentracijah izvedli indirektno gojitveno metodo štetja glive Candida albicans na diferencialnem gojišču HiCrome Ogye agar. Pri vseh koncentracijah smo tudi izmerili absorbanco pri valovnih dolžinah med 420 nm in 680 nm in izdelali umeritvene krivulje za vsako valovno dolžino. Rezultati Iz umeritvenih krivulj smo določili korelacijski koeficient R2. Pri 560 nm smo dobili rezultat R2= 0,989 in se najbolj približali številu R2=1. Največje odstopanje smo dobili pri umeritveni krivulji za določanje koncentracije glive Candida albicans pri 430 nm, kjer smo dobili rezultat R2= 0,6822. Diskusija in zaključki Ugotovili smo, da je indirektna metoda štetja glive Candida albicans z gojitvijo na diferencialnem gojišču HiCrome Ogye agarju primerljiva z merjenjem absorbance tekočega gojišča s pomočjo spektrofotometra pri valovni dolžini 560 nm.Candida albicans is one of the many saprophytic fungi on the human mucosa. It is found in the mouth, respiratory system, gastrointestinal tract and vagina of 25% of healthy population. It is the most common fungus that is responsible for most of localized fungal infections in humans. Patients with reduced immune resistance, oncology patients and patients infected with HIV are particularly at risk for the development of the fungus Candida albicans. Methods In this study the indirect cultivation method for counting the fungus Candida albicans on the differentiated medium HiCrome Ogye agar at different concentrations was conducted. At all concentrations, we also measured the absorbance at wavelengths between 420 nm and 680 nm and prepared calibration curves for each wavelength. Results The correlation coefficient R2 was determined from each of the calibration curves. At 560 nm we achieved the result of R2 = 0.989 which was the closest to the number R2 = 1. The maximum deviation was obtained from the calibration curve for determining the concentration of the fungus Candida albicans at 430 nm, where we get the result of R2 = 0.6822. Discussion and conclusions: We have found that the indirect cultivation method of counting the fungus Candida albicans on the differential medium HiCrome Ogye agar comparable to measuring the absorbance of the liquid culture medium by means of a spectrophotometer at the wavelength of 560 nm.
