Abstract     Abstract. Aim of the study: We used DMEM plant extracts of Capsicum chinense and Allium sativum, in previous studies where we determined that cellobiose, sulphur, selenium and vitamin C are the main effective molecules for the onset of programmed cell death in various types of cancer and immortalized cells. This study aims to elucidate the effect of cellobiose, sulphur, selenium and vitamin C isolated by HPLC from the combination of two cold plant extracts of Al[!]ium sativum and Capsicum chinense on the onset of programmed cell death in primary MEF wt and immortalized MEF wt cells. In continuation these data serves us as the platform for programming computerized virtual animal and human model in order to investigate, whether the effect of above mentioned combination could be compared to oxilaplatin results. Materials and methods: For determination of cell death by the mixture of two DMEM cold plant extracts and unique molecule complex cellobiose, sulphur, selenium and vitamin C we used MTS assay, caspase-3 activation test and staining with annexin and 7-AAD. To investigate how cell death is executed, we used dyes to study organelle integrity in our case Mito Tracker Red CMXRos to detest the integrity of outer membrane potential of mitochondria. Results: The present study confirmed that the unique combination of cellobiose, sulphur, selenium and vitamin C in the concentration range from 1ug/mL significantly increased the apoptotic and necrotic cell number, caspase-3 activation and loss of mitochondrial outer membrane potential only in immortalized cell line MEF wt. Conclusion: Combination of cellobiose, selenium, sluphur and vitamin C in comparable concentrations as the plant extract mixture induce programmed cell death in MEF wt immortalized cell line, but not in primary cell line MEF wt. Only solution of cellobiose in comparable concentrations to plant extract mixture is highly not enough to trigger programmed cell death. With our virtual animal and human model for medicine testing we discovered this combination has comparable effect as oxaliplatin differing only in the concentrations used.
