Abstract     Background. For studying intracellular multiplication of bacteria Legionella pneumophila, the cell culture of alveolar macrophages is usualy used. The purpose of our study was to find out, if Legionella pneumophila can multiplicate in nonphagocytic cell culture. On this model, the influence of human alpfa interferon (IFN) on cells, infected with bacteria Legionella pneumophila, was studied. Methods. At first, bacteria Legionella pneumophila was cultivated on WiREF cells. An infective dose of bacteria, able to cause plaques, was established. The plaques were counted and it was found out that the number of plaques in cell culture depend of the bacterial inoculum. Also, when staind with methylene blue, the amount of eluated coulour was in correlation with the number of plaques. Results. It was found that the cells which were pretreated with interferon alpha developed the "antibacterial state", which is analogous to the antiviral state and they were resistant to the infection with the bacteria Legionella pneumophila. Conclusions. The developed test model has the advantage, that it is repeatable and could be usefull for testing the influence of, for example, antibiotics on Legionella pneumophila.
Summary     Izhodišča. Za študij znotrajceličnega razmnoževanja bakterije Legionella pneumophila le-to navadno gojimo v celični kulturi alveolarnih makrofagov. Namen naših poskusov je bil ugotoviti, ali se bakterija Legionella pneumophila lahko razmnožuje v nefagocitni celični kulturi WiREF. V tem testnem sistemu smo preučevali vpliv humanega alfa interferona (IFN) na celice, inficirane z bakterijo Legionella pneumophila. Metode. Z bakterijo Legionella pneumophila smo okužili celično kulturo WiREF in ugotovili, da v njej povzroča citopatske spremembe-plake. Le-te smo šteli in ugotovili odvisnost med številom plakov na celicah in bakterijskim inokulumom. Pri barvanju celic z barvilom metilensko modro (MM) smo ugotovili, da količina barvila eluiranega iz celic, korelira s številom plakov. Rezultati. Dokazali smo, da je to bakterijo mogoče gojiti v celicah WiREF in ugotovili infektivni odmerek za nastanek plakov. Ugotovili smo, da poprejšnja obdelava celic z alfo interferonom povzroči nastanek "protibakterijskega stanja" celic WiREF, ki so postale bolj odporne za kasnejšo okužbo z bakterijo Legionella pneumophila. Zaključki. Testni model, ki smo ga razvili, ima to prednost, da so testi ponovljivi in bi bil primeren za testiranje delovanja antibiotikov na bakterijo Legionella pneumophila.
