Abstract     Backround. The clinical uses of interferons and other cytokines employ the subcutaneous, intramuscular, intravenous, or intratumoral administration. This study investigates, whether some cytokines (interferon alpha, interleukin-2 and tumor necrosis factor) exert systemic effects following their oral administration to mice. Methods. Experimental mice received oral and subcutaneous cytokine treatment. After three days of treatment the number of peripheral leukocytes and bone marrow colonies was determined. Results. Oral administration of interferon alpha and interleukin-2 was found to cause a significant suppression of peripheral leukocyte counts and bone marrow proliferative activity. The levels of suppression were similiar to those seen with subcutaneously administered interferon alpha and interleukin-2. Conclusions. The results demonstrate that orally administered interferon alpha and interleukin-2 can exert systemic effects in mice. They further suggest a potential clinical role for oral administration of cytokines.
Summary     Izhodišče. Interferon, interlevkin in druge citokine običajno bolnikom vbrizgavajo pod kožo, v mišico, v žilo ali v tumor. V pričujoči raziskavi smo poskušali ugotoviti, ali nekateri citokini (interferon alfa, interlevkin-2, tumor nekrotizirajoči faktor) po oralni aplikaciji povzročajo sistemske učinke v poskusnih miših. Metode. Poskusnim mišim smo dajali skozi usta ali podkožno različne koncentracije citokinov. Po treh dneh smo določili število levkocitov v periferni krvi in število granulocitno-makrofagnih kolonij v kultiviranem kostnem mozgu. Rezultati. Oralna aplikacija interferona alfa in interlevkina-2 je povzročila signifikantno zmanjšanje števila levkocitov in mieloidnih kolonij kostnega mozga. Stopnja zaviranja je bila podobna kot pri podkožni aplikaciji interferona alfa in interlevkina-2. Zaključki. Rezultati kažejo, da oralna aplikacija interferona alfa in interlevkina-2 povzroča sistemske učinke v miših. To nakazuje, da bi mogoče lahko preizkusili oralno aplikacijo citokinov tudi v kliničnih študijah.
