Abstract     Background. The commercially available polymerase chain reaction (PCR) based assay, Amplicor Chlamydia Trachomatis Test, for the detection of Chlamydia trachomatis DNA in patients with urogenital infections was compared with cell culture of Clamydia trachomatis and direct fluorescent antibody (DFA) assay. Methods. Two groups of patients were included in the study: 13 male, attending at the Clinic for Dermatology and 15 female, attending at the Clinic for Gynaecology and Obstetrics in Ljubljana. PCR method was used to test urine and sample from the urethra in male, and from the cervix in female. Male urethral specimens and cervical specimens were tested by cell culture and male urethral specimens also by DFA staining. Results. Overall, 41 samples from the urethra, cervix and urine were tested with cell culture, DIF and PCR methods. Discrepant results have been found in two male. Conclusions. The commercially available test Amplicor Clamydia Trachomatis Test revealed to be a suitable method of choice for detection of chlamydial infection, particularly when there are discrepancies in the results of standard assays. By our preliminary results we showed that urine specimen is suitable sample for detection of C. trachomatis DNA in male with urogenital infections.
Summary     Izhodišča. Z molekularno metodo verižne rakcije s polimerazo (PCR, polymerase chain reaction) smo želeli dokazati DNA Chlamydia trachomatis v vzorcih bolnikov z urogenitalnimi okužbami. Pri tem smo primerjali komercialni test za izvedbo PCR, Amplicor Chlamydia trachomatis Test, z metodo izolacije C. trachomatis v celični kulturi in z metodo neposredne imunofluorescence (DIF). Metode. V študijo smo vključili dve skupini preiskovancev: 13 moških, ki so se zdravili ambulantno na Dermatološki kliniki, in 15 žensk, ki so iskale pomoč na Ginekološki kliniki v Ljubljani. Z metodo PCR smo preiskovali vzorce urina in kužnine iz uretre pri moških ter kužnine iz cerviksa pri ženskah. Izolacijo C. trachomatis smo naredili iz kužnin iz uretre pri moških in iz kužnin iz cerviksa pri ženskah, test DIF pa le iz brisov uretre pri moških. Rezultati. Z metodo izolacije v celični kulturi, DIF in PCR smo skupno testirali 41 kužnin uretre, cerviksa in urina. Do neujemanja rezultatov je prišlo pri dveh bolnikih. Zaključki. Primerjalna študija je pokazala, da je komercialno dostopen test Amplicor Chlamydia trachomatis test primerna metoda izbora predvsem takrat, ko pride do razhajanj v rezultatih standardnih postopkov odkrivanja klamidijskih okužb. Preliminarni rezultati so potrdili, da je urin ustrezna kužnina za dokaz DNA C. trachomatis z metodo PCR pri moških z urogenitalnimi okužbami.
