Abstract     In isolated rat hearts verapamil effects on partial ishaemia (ligature of the anterior interventricular artery) and global hypoxia (perfusate saturated with nitrogen) were compared. Verapamil (20 nmol L-1) was added to the perfusate either 10 minutes before the beginning, or simultaneously with the beginning of partial ischameia or global hypoxia. The degree and extent of injury were assessed by the release rate of lactate dehydrogenase in venous effluents, by coronary flow, contractility, heart rate and ECG. In both types of injury verapamil showed a cytoprotective effect which was expressed especially upon termination of ischamia and hypoxia, i.e. during reperfusion and reoxygenation, respectively. During reperfusion verapamil was effective regardless of whether applied before or at the beginning of ischaemia, while during reoxygenation it was markedly more effective if given before hypoxia. In verapamil-pretreated hearts verapamil proved more efficient in more parameters in hypoxia + reoxygenation than in ischaemia + reperfusion group.
