Abstract | Background. The reticulocytes represent the penultimate maturation phase of erythroid cells. The reticulocyte count is a good index of erythropoetic activity. Commonly used microscopic counting technique is time-consuming and the precision is poor. Recently flow cytometric reticulocyte analysis has been developed and today it is becoming an increasingly popular method for quantification of reticulocytes. These can be accomplished by using thioflavin T, pyronin Y, acridine orange, ethidium bromide, thiazole orange or auramine orange. Conclusions. This review will recount anatomy and physiology of reticulocytes, the identification and counting of these cells with different methods, mainly with flow cytometry. | |
Summary | Izhodišča. Retikulociti so celice rdeče vrste v končnem obdobju dozorevanja. Določitev števila retikulocitov v krvi je indirekten način za oceno obsega nastajanja eritrocitov v kostnem mozgu. Štetje retikulocitov s svetlobnim mikroskopom je zamudno in nenatančno. Danes se uveljavljajo načini določanja števila retikulocitov s pretočnim citometrom. To omogočajo fluorescenčna barvila kot so tioflavin T, pironin Y, akridin oranž, etidium bromid, tiazol oranž ali auramin oranž. Zaključki. V tem prispevku je pregled zgradbe in razvoja retikulocitov, prepoznavanje in načini določanja števila retikulocitov s povdarkom na uporabi pretočnega citometra. | |
