Abstract     Background: Hardness is one of the key biomechanical properties of mineralized dental tissues. Our aim was to determine hardness of the dentine forming roots of primary and permanent teeth at a measured distance of hardness test from the cemento-dentinal junction (CDJ). Methods: A sample of 11 primary and 14 permanent caries free mandibular incisors was selected from collection of extracted teeth. Roots were sectioned transversely below the cemento-enamel junction, embedded in epoxy resin and polished metalographically. Vickers hardness was determined with a universal indenter (Shimadzu, Kyoto, Japan) by applying 100g force for 10 seconds. Measurements were made at regular intervals from the CDJ to the root canal in the middle of each root surface. A Student's t-test for independent samples and one-way ANOVA followed by Tukey's (HSD) procedure were used to analyze the data. Results: On average, the root dentine was harder in permanent than in primary teeth (68.9 HV vs. 62.1 HV; p < 0.001). In both groups of teeth, hardness of the dentine decreased with increasing distance of hardness test from the CDJ. In permanent teeth, the dentine was harder on approximal surfaces (mesially 71.0 HV, distally 74.1 HV) than on labial (67.1 HV) and lingual surfaces (66.0 HV) (p < 0.001). Conclusions: Hardness of the root dentine decreases in pulp direction, whereby the outer, middle, and inner thirds of the dentine wall are harder in permanent than in primary teeth. In roots of permanent teeth, the dentin on approximal surfaces is harder than on other root surfaces.
