Abstract     We have generated MLi003-A, a new induced pluripotent stem cell (iPSC) line derived from hair follicle kera-tinocytes of a healthy male characterized with a maximum number offilaggrin tandem repeats, making this iPSCline the best control for studies on skin barrier function. The characterization of the MLi003-A cell line consistedof molecular karyotyping, high-throughput array-based sequencing composed of Fluidigm microfluidics tech-nology and next-generation sequencing of thefilaggrin alleles, and pluripotency and differentiation potentialstesting by immunofluorescence of associated markers bothin vitroandin vivo. The MLi-003A line has been alsotested for ability to differentiate into keratinocytes.
