Title     Effect of farnesol on 3-hydroxy-3-methylglutaryl CoA reductase activity in semi-intact CHO-215 cells

Abstract     A model of CHO-215 semi-intact cells was used for the study of the regulatory mechanisms of HMGCoA reductase at the postranslational level. In such a system by removing a portion of the plasma membrane large perforations are created that allow the release of soluble cytoplasmic components but leave the intracellular organelles intact. The short term effect of cholesterol and non-sterol intermediates of the isoprenoid pathway on HMGCoA reductase activity was stidied by monitoring enzyme activities of solubilized enzyme and intact enzyme in situ. Elevated levels of cholesterol always resulted in a decrease of enzyme activity in semi-intact CHO-215 independendt of the way that enzyme activity, was measured. Among non-sterol intermediates of the isoprenoid pathway, famesol was found to increase the HMGCoA reductase activity.
