Abstract | The polymerase chain reaction (PCR) is an in vitro method which uses enzymatic synthesis to amplify, in an exponential manner, specific DNA or RNA sequences. Although it has been available for only a few years, PCR technology is already finding widespread application in biomedical research. This article presents a broad overview of the principles of PCR, sumarizes some of the application problems, and described some measures taken to overcome them. The main applications of PCR-based assays in virology are also discussed. | |
Summary | Verižna reakcija s polimerazo (PCR) je razmeroma nova metoda sinteze nukleinskih kislin in vitro, s katero lahko v kratkem času pomnožimo določeni odsek DNA v velikem številu kopij. Čeprav je metoda stara šele nekaj let, je danes že nepogrešljiva na številnih področjih biomedicinskih raziskav. Namen našega članka je predstaviti tako teoretične osnove PCR in načine optimizacije metode kot tudi najbolj pogoste probleme in težave, ki se pojavljajo pri izvedbi PCR, ter ukrepe za njihovo odpravo oz. zmanjšanje. Na kratko so prikazane tudi današnje možnosti uporabe PCR v raziskovanju in diagnostiki virusnih okužb. | |
