Abstract     Despite numerous laboratory tests, a full case history clinical status still remain the pillars of diagnosis in patients with suspect immunologic or rheumatologic disease. Numerous tests exist which can aid us in setting the diagnosis. In clinical immunology and rheumatology a target-specific diagnosis is of extreme importance, due above all to the high costs. Laboratory investigations are oriented towards the differentiation between primary and secondary immunodeficiency, structural and functional immunodeficiency, humoral immunodeficiency, a defect of T lymphocytes, granulocytes and complement as well as towards the diagnosis of autoantibodies. The aim of the present paper is to present some of them as well as their significance in the setting the diagnosis.
