Abstract     Carbapenems are broad spectrum antibiotics possessing potent antibacterial activities against Gram-positive and Gram-negative bacteria, some of them also exhibit inhibitory activity against bacterial beta-lactamases. Thienamycin produced by Streptomyces cattleya represents a lead compound for development of new carbapenem antibiotics. The absolute configuration at positions 5, 6 and 8 of carbapenem nucleus influences the antibacterial activity, stability and also inhibitory activity against bacterial beta-lactamases. Introduction of 1-beta-methyl substituent to the carbapenem nucleus increases metabolical stability (stability against renal dehydropeptidase-1) and antibacterial activity, including activity against Pseudomonas sp. and MRSA in some cases.
