Abstract     To provide a colloidal carrier system loaded with zinc-phthalocyanine (ZnPc) for photodynamic therapy biodegradable poly(D,L-lactic acid) (PLA) nanoparticles were prepared by the salting-out method and solvent diffusion method. For appropriate nanoparticle features and loading capacity, the PLA concentration in the organic phase, the protective hydrocolloid concentration in the aqueous phase, and the organic/aqueous phase ratio played a predominant role. The nanoparticle formation was dependent on the rate of diffusion of the organic solvent into the aqueous phase, thus influencing polymer precipitation. The loading capacity and entrapment efficiency were high. ZnPc-loaded PLA nanoparticles were also purified and freeze-dried. In order to obtain well redispersible PLA nanoparticles, the presence of additives such as trehalose, lactose or glucose was of great importance; the freezing temperature played a minor role. PLA nanoparticles have a regular spherical shape with a mean diameter of 300 nm or less.
