Abstract     The object of this study was to determine reference levels of vitamin D and its metabolites in the sera of individuals with normal vitamin D metabolism. The measurements were done in compliance with the instructions for reference value determinations given by International Federation of Clinical Chemistry. The paper presents the nomenclature, physical and chemical properties, and physiology and pathologic physiology of vitamin D, as well as various diseases associated with changed serum levels of vitamin D and its metabolites. There follow the description of the method for determining vitamin D levels, used at the Institute of Clinical Chemistry and Clinical Biochemistry, which allows for simultaneous determinations of three vitamin D metabolites and consists of the following three phases: serum deproteinization (acetonitrile), solid phase extraction, and high pressure liquid chromatography and competitive protein binding assay. Reference individuals for our study were selected prospectively: we used blood samples obtained from them on periodic medical examinations. After excluding samples which failed to meet the set criteria of vitamin D physiology and pathologic physiology, we obtained 120 samples for calciol and calcitrol determinations, and 240 samples for calcidiol measurements. The latter were further divided into a spring and autumn subgroup. The results were analysed using a nonparametric statistical test. In addition a normal metabolite distribution (Gauss) was applies. We obtained mean 0.95 intervals with a 0.90 confidence interval for the reference values of calciol, calcidiol and calcitriol of adult males in Slovenia, considered healthy according to their vitamin D metabolism.
Summary     V predstavljenem delu smo določili orientacijske referenčne vrednosti vitamina D in njegovih presnovkov v krvnem serumu ljudi, ki jih imamo glede na presnovo vitamina D za zdrave. Pri delu smo sledili priporočilom, ki jih je za določanje referenčnih vrednosti objavila Mednarodna zveza za klinično kemijo. Najprej smo predstavili nomenklaturo, fizikalno-kemijske lastnosti, fiziologijo in patofiziologijo vitamina D ter najznačilnejše bolezni, povezane s spremembo serumskih koncentracij vitamina D in presnovkov. V nadaljevanju smo opisali analizni postopek za določanje koncentracije vitamina D in presnovkov, ki ga uporabljamo na Inštitutu za klinično kemijo in klinično biokemijo. Izbrali smo metodo, kjer vse tri presnovke določamo sočasno. Metoda ima tri stopnje: deproteinizacija seruma (acetonitril), ločevanje presnovkov in določanje koncentracije posameznih presnovkov. Referenčne posameznike smo izbrali prospektivno - uporabili smo kri, ki je ostala po analizah ob rednih sistematskih pregledih. Po izključevanju na podlagi meril, ki smo jih postavili glede na ugotovitve o fiziologiji in patofiziologiji vitamina D, smo dobili 120 vzorcev za kalciol in kalcitriol, za kalcidiol pa smo imeli na razpolago 240 vzorcev, ki smo jih razdelili na spomladansko in jesensko podskupino. Izbrali smo neparametrično statistično obravnavo, kljub temu pa smo določili tip porazdelitve, ki je bila pri vseh presnovkih normalna (Gaussova). Nato smo izračunali range 2,5 in 97,5 percentila. Te predstavIjajo spodnjo in zgornjo mejo referenčnega intervala. Nato smo izračunali še intervale zaupanja 0,90 za zgornje in spodnje meje referenčnega intervala. Ugotovili smo tudi, da se spomladanske in jesenske vrednosti kalcidiola med seboj značilno razlikujejo. (Abstract truncated at 2000 characters).
