Abstract     Histochemical, immunohistochemical and ultrastructural methods are very important in the study of qualitative composition of skeletal muscles. The research was performed on samples of Longissimus dorsi (Ld) and Semimembranosus (Sm) muscles of Chianina breed of 12 months old steers. For the histochemical and immunohistochemical investigations samples of muscle were frozen in isopentane previously cooled in liquid nitrogen. Serial sections were processed to demonstrate NADH-tetrazolium reductase, alpha- glycerophosphate dehydrogenase and myosinic ATPase activity, so it's possible distinguish different types of skeletal muscular fibres; immunohistochemical technique were used to evaluate the percentage of connective tissue. The slides were examined by an image analyzer. Ultrastructure investigations were performed on specimens of 0,5 mm3 fixed with Karnowsky's solution and embedded in epoxy resin. The sections, observed at T.E.M., provided indications about some ultrastructural aspects of skeletal muscular fibres, particularly to evaluate the percentage of "splitting" myofibrils.
