Abstract | Nine hundred and fourteen adult saws of three different breeds were tested for the ryanodine receptor 1 (RYR 1 ) gene mutation. After the isolation of the genomic DNA a polymerase chain reaction (PCR) was used to amplify the DNA fragment of the gene which was digested with a Cfo I restriction endonuclease. Point mutation was detected on the agarose gel electrophoresis. Three different genotypes were identified inside each breed: dominant homozygotes (NN), heterozygotes (Nn) and recessive homozygotes (nn). Out of 914 saws tested, 36.11 % were heterozygotes and 7.00% homozygotes for the RYR 1 gene mutation. The mutated RYR 1 gene frequency in the population of all three breeds is 0.25055. The highest percent of recessive homozygotes (nn) (over 18%) and also the highest percent of heterozygotes (Nn) (over 56%) was detected in the German Landrace breed. In the Large White and Swedish Landrace there were around 25% heterozygotes and less than one percent were recessive homozygous saws. The difference in the incidence of the RYR 1 gene mutation was highly significant between the German Landrace and the other two breeds (Swedish Landrace, Large White); no significant differences were observed between Swedish Landrace and Large White breeds. | |
