Abstract | Malignant lymphomas are neoplasms characterized by proliferation of cells derived from the lymphoid tissue and its precursors. Clonality of lymphoid proliferations is of important diagnostic value for distinguishing between neoplastic lesions and reactive processes. The majority of lymphoid malignancies are of monoclonal origin while reactive proliferations are of polyclonal origin, although clonality does not necessarily correlate with a clinically malignant course of the disease. Determination of clonality using molecular biological techniques is based on the process of rearrangement in the genes coding for B and T cell antigen receptors. These rearranged gene sequences can be amplified by the polymerase chain reaction (PCR) and electrophoretically separated in order to determine the clonality. We introduced the method for clonality analysis of lymphoprolifeartive disorders using the polymerase chain reaction. We analysed 20 lymphoid proliferations of B cell origin and 30 of T cell origin as well as several control samples using the PCR method. We determined the clonality of all analysed B cell proliferations and 22 of 30 analysed T cell proliferations. The method is relatively simple, quick and can be performed on formalin-fixed, paraffin-embedded tissue. Taking into consideration described characteristics it can be used as additional method for routine diagnosis of lymphoproliferative disorders in the Department of Pathology of the Institute of Oncology in Ljubljana. | |
