Abstract     Lyme borreliosis is the most common tick-borne disease in the northern hemisphere. in Slovenia, Lyme borreliosis is endemic. Signs and symptoms of Lyme borreliosis vary, the most common clinical manifestation following the tick bite is skin rash, erythema migrans. Disseminated infection includes severe neurologic, skin, cardiac and arthritic manifestations, all registered in Slovenia. The etiologic agent of Lyme borreliosis, Borrelia burgdorferi sensu lato, has been isolated world-wide from various species of ixodid ticks and from animal and human material. Initially identified as one species, B. burgdorferi sensu lato has recently been in respect to immunological and molecular heterogeneity delineated into three pathogenic species: B. afzelli, B. garini and B. burgdorferi sensu stricto. The aim of the present study was to assess the phenotypic and genotypic diversity of a large number of B.burgdorferi sensu lato strains isolated from human material in Slovenia. Skin biopsies, CSF and blood from patients with different clinical manifestations of Lyme borreliosis have been the source of our strains. Borreliae were cultivated in MKP medium at 33 degrees C. We analised 233 strains. Sodium-dodecyl sulphate polyacrylamide gel electrophoresis was performed for protein separation. Borrelial DNA was prepared by gel insert method. For plasmid detection, pulsed field gel electrophoresis (PEGE) was performed. For species identification, DNA was additionally digested by MluI restrictin enzyme and DNA fragments were separated by PEGE. We analysed species of 112 strains: 92 (82%) were B.afzelli, 19 (17,1%) B.garinii and only one (0,9%) B.burgdorferi sensu stricto. We analysed plasmid profiles of 63 isolates: 62 among them had alarge plasmid in the range of 49-63kb, and 3 to 7 plasmids in the range of 15 to 38kb.(Abstract truncated at 2000 characters.)
